Start by marking the mating ends of your workpieces and labeling each as to whether it is a tail board or a pin board. When hand-cutting dovetails, we typically cut the tails first and then use those to lay out the pins. Measure the thickness of the board that will receive the pins, and then set your marking gauge to match this thickness. Scribe a line all the way around the tail board at each end.
